New VITROCELL® Spiking System for the generation of gas mixtures in turnkey in vitro exposure systems on IUTOX 2010.

VITROCELL® presented a new solution for the exposure of cells or bacteria to gas mixtures and volatile organic compounds (VOC) at the 2010 IUTOX conference in Barcelona / Spain from 19. ? 23. July 2010.

(PresseBox) - The VITROCELL® Spiking System (VSS) offers an excellent possibility to prepare gas mixtures from solvents, gases and vapours in the ppm and ppb ranges. It is a precise solution when the required components and concentrations are not available in cylinders.
The gas mixture is produced dynamically by means of a digital syringe pump allowing injections of less than 0.1 µl with high accuracy. The component is introduced into a zero gas. Programmable heating steps, mass flow controllers and the secondary dilution system allow the production of mixtures in broad concentration ranges.
Digital Syringe Drive
The microprocessor controlled syringe drive is compatible with a large range of commercially available syringes from 10 µl to 140 ml. The selection is made out of an internal syringe library. The flow rate and operation time can be defined. The injector block can be easily adjusted to fit for the diameters of different syringe types. The injector block is heated in function of the dew point of the component. Further heating takes place in the subsequent heating line.
Dilution / Distribution System
The secondary dilution system dilutes the mixture further to the ppb range. It makes use of the proven VITROCELL® double jet technology. It also acts as a distribution system as it is connected to the VITROCELL® exposure module.
Turnkey installations for the analysis of gases and complex mixtures
The analysis of gases, volatile organic compounds (VOC) and complex mixtures from combustion processes such as cigarette smoke or Diesel exhaust is performed with special configurations. All installations are delivered turnkey including training.
About the VITROCELL® cell cultivation and exposure technology for gases, complex mixtures and nano particles
The direct exposure of the cells takes place at the air liquid interface in the VITROCELL® cell cultivation and exposure modules. For this purpose, the cells are first cultivated on membrane inserts (e. g. ThinCerts® from Greiner, Costar® from Corning or Falcon® from Becton Dickenson). The membrane inserts are placed in the modules which are tempered at 37°C. The medium supply is performed either statically or by permanent exchange. In both cases the cells receive the cell culture medium from below through the membrane of the insert.




In the direct exposure technology, the test atmosphere is delivered to the cells via specially treated aerosol inlets using a low vacuum flow. As the cells are not covered with medium, they are exposed most efficiently at the air liquid interface and can therefore react in an optimal manner. After exposure, the membrane inserts are taken out of the module and cells are forwarded to standard routines for endpoint analysis.
The VITROCELL® product range helps to reduce animal testing in the field of e. g. inhalation toxicology. For this purpose human cells are cultivated in special exposure modules on membranes and then exposed at the air/liquid interface to the test substances. The same technology can also be applied to bacteria.
